重组腺病毒介导的1.2拷贝HBV DNA3种肝细胞中表达的研究


Expression of HBV in three hepatocytes by 1.2-copy-HBV DNA mediated by recombinant adenovirus

Abstract: Objective To study the HBV expression in three hepatocytes infected with recombinant adenovirus which containing 1.2 copies HBV DNA. Methods One chicken hepatoma cell and two types of human hepatocytes were infected by the recombinant adenovirus at 25 pfu / cell. HBV specific mRNA was detected by RT-PCR after 3 days infected, the HBsAg and HBeAg was detected by ELISA and HBV DNA by real time PCR every day after infected. Results HBV mRNA was detectable in these three hepatocytes after tranfected with the recombinant adenovirus. The quantities of HBV DNA and HBV antigens were increased by the time. Conclusions HBV infectious state could be established in these cells after infected by recombinant adenovirus.

乙型肝炎病毒宿主范围很窄,感染具有高度的种属和组织特异性,至今尚无成熟的体外细胞培养模型,因此,研究HBV的生物学特性必须借助于一定的基因转移系统,将HBV导入靶细胞,使其在细胞内获得表达。腺病毒是目前最具应用潜力的载体之一,其能利用病毒自身感染宿主细胞的特点,简便有效地将目的基因导入靶细胞,并使其有效表达[1,2]。我们曾利用腺病毒的泛嗜性,将重组腺病毒感染人肝癌细胞HepG2,获得能表达HBV标志cccDNA的细胞模型[3]。然而对于不同来源的肝源化细胞,重组腺病毒介导的HBV是否具有相同的表达效率,还需进一步研究。本文利用L02、HepG2和LMH3种不同肝源化细胞株,观察HBV在不同细胞中的表达情况,以进一步了解该重组HBV腺病毒的生物学特性。
